    NEW 3D METHOD TO ESTIMATE THE CYCLING FRONTAL AREA DURING PEDALLING

    Aerodynamic drag represents a high percentage of the resistive forces in cycling. The frontal area has been traditionally measured through 2D digitalization, which requires a capture and an analysis processes. In the present study, the 2D method has been compared with a new real time 3D method to measure the frontal area in three different cycling positions. High reliability (ICC\u3e0.9) was found on both measurement methods. However there were differences between methods at the most aerodynamic positions. These differences might appear due to the capacity of the new method to measure the depth dimension, which supposes an improvement on the frontal area analysis.     The consequences of a year of the COVID-19 pandemic for the mental health of young adult twins in England and Wales

    Background The COVID-19 pandemic has affected all our lives, not only through the infection itself but also through the measures taken to control the spread of the virus (e.g. lockdown). Aims Here, we investigated how the COVID-19 pandemic and unprecedented lockdown affected the mental health of young adults in England and Wales. Method We compared the mental health symptoms of up to 4773 twins in their mid-20s in 2018 prior to the COVID-19 pandemic (T1) and during four-wave longitudinal data collection during the pandemic in April, July and October 2020, and in March 2021 (T2-T5) using phenotypic and genetic longitudinal designs. Results The average changes in mental health were small to medium and mainly occurred from T1 to T2 (average Cohen d = 0.14). Despite the expectation of catastrophic effects of the pandemic on mental health, we did not observe trends in worsening mental health during the pandemic (T3-T5). Young people with pre-existing mental health problems were disproportionately affected at the beginning of the pandemic, but their increased problems largely subsided as the pandemic persisted. Twin analyses indicated that the aetiology of individual differences in mental health symptoms did not change during the lockdown (average heritability 33%); the average genetic correlation between T1 and T2-T5 was 0.95, indicating that genetic effects before the pandemic were substantially correlated with genetic effects up to a year later. Conclusions We conclude that on average the mental health of young adults in England and Wales has been remarkably resilient to the effects of the pandemic and associated lockdown

    The role of APOBEC3B in lung tumor evolution and targeted cancer therapy resistance

    In this study, the impact of the apolipoprotein B mRNA-editing catalytic subunit-like (APOBEC) enzyme APOBEC3B (A3B) on epidermal growth factor receptor (EGFR)-driven lung cancer was assessed. A3B expression in EGFR mutant (EGFRmut) non-small-cell lung cancer (NSCLC) mouse models constrained tumorigenesis, while A3B expression in tumors treated with EGFR-targeted cancer therapy was associated with treatment resistance. Analyses of human NSCLC models treated with EGFR-targeted therapy showed upregulation of A3B and revealed therapy-induced activation of nuclear factor kappa B (NF-κB) as an inducer of A3B expression. Significantly reduced viability was observed with A3B deficiency, and A3B was required for the enrichment of APOBEC mutation signatures, in targeted therapy-treated human NSCLC preclinical models. Upregulation of A3B was confirmed in patients with NSCLC treated with EGFR-targeted therapy. This study uncovers the multifaceted roles of A3B in NSCLC and identifies A3B as a potential target for more durable responses to targeted cancer therapy.</p

    The genetic architecture of the human cerebral cortex

    The cerebral cortex underlies our complex cognitive capabilities, yet little is known about the specific genetic loci that influence human cortical structure. To identify genetic variants that affect cortical structure, we conducted a genome-wide association meta-analysis of brain magnetic resonance imaging data from 51,665 individuals. We analyzed the surface area and average thickness of the whole cortex and 34 regions with known functional specializations. We identified 199 significant loci and found significant enrichment for loci influencing total surface area within regulatory elements that are active during prenatal cortical development, supporting the radial unit hypothesis. Loci that affect regional surface area cluster near genes in Wnt signaling pathways, which influence progenitor expansion and areal identity.     Three-dimensional tibiofemoral kinematics of the anterior cruciate ligament-deficient and reconstructed knee during walking

    Background: It is possible that gait abnormalities may play a role in the pathogenesis of meniscal or chondral injury as well as osteoarthritis of the knee in patients with anterior cruciate ligament deficiency. Hypothesis: The three-dimensional kinematics of anterior cruciate ligament-deficient knees are changed even during low-stress activities, such as walking, but can be restored by reconstruction. Study Design: Case control study. Methods: Using a three-dimensional optoelectronic gait analysis system, we examined 13 patients with anterior cruciate ligament-deficient knees, 21 patients with anterior cruciate ligament-reconstructed knees, and 10 control subjects with uninjured knees during walking. Results: Normal patterns of knee flexion-extension, abduction-adduction, and internal-external rotation during the gait cycle were maintained by all subjects. A significant difference in tibial rotation angle during the initial swing phase was found in anterior cruciate ligament-deficient knees compared with reconstructed and control knees. The patients with anterior cruciate ligament-deficient knees rotated the tibia internally during the initial swing phase, whereas the others rotated externally. Conclusions: Patients with anterior cruciate ligament-deficient knees experienced repeated episodes of rotational instability during walking, whereas patients with reconstruction experienced tibial rotation that is closer to normal.     How metaphors influence semantic relatedness judgments: the role of the right frontal cortex

    We used event-related fMRI (ER-fMRI) to test the hypothesis that metaphors bias cognitive processing of semantic relatedness towards a search for a wider range of associations. Twelve right-handed male volunteers read a mixture of metaphoric and literal sentences, each sentence being followed by a single word, which could be semantically related or not to the preceding sentence context. We found that judging unrelated words as contextually irrelevant was associated with increased blood oxygenation level-dependent (BOLD) signal in the right ventrolateral prefrontal cortex in the metaphoric but not in the literal condition. The same region was also activated when subjects endorsed a semantic relation between words and metaphoric sentence primes but not between words and literal sentence primes. We argue that these results are consistent with the notion of semantic open-endedness, whereby figurative statements bias cognitive processing towards a search for a wider range of semantic relationships compared to literal statements, and thus lend further support to the view that coarse semantic coding occurs preferentially in the right hemisphere
